Arthritis is a form of joint disorder that involves inflammation of one or more joints. Osteoarthritis (OA) also known as degenerative arthritis or degenerative joint disease or osteoarthrosis, is a group of mechanical abnormalities involving degradation of joints including articular cartilage and subchondral bone. Symptoms may include joint pain, tenderness, stiffness, locking, and sometimes an effusion. A variety of causes—hereditary, developmental, metabolic, and mechanical deficits—may initiate processes leading to loss of cartilage. When bone surfaces become less well protected by cartilage, bone may be exposed and damaged. As a result of decreased movement secondary to pain, regional muscles atrophy, and ligaments may become more lax.


  • Pain - Your joints may ache, or the pain may feel burning or sharp. For some people, the pain may come and go. Constant pain or pain while you sleep may be a sign that your arthritis is getting worse.
  • Stiffness - When you have arthritis, getting up in the morning can be hard. Your joints may feel stiff and creaky for a short time, until you get moving. You may also get stiff from sitting.
  • Muscle weakness - The muscles around the joint may get weaker. This happens a lot with arthritis in the knee.
  • Swelling - Arthritis can cause swelling in joints, making them feel tender and sore.
  • Deformed joints - Joints can start to look like they are the wrong shape, especially as arthritis gets worse.

Rheumatoid arthritis (RA) is an autoimmune disease that results in a chronic, systemic inflammatory disorder that may affect many tissues and organs, but principally attacks flexible (synovial) joints. It can be a disabling and painful condition, which can lead to substantial loss of functioning and mobility if not adequately treated.


  • Stiffness - The joint is harder to use and might have a limited range of motion. "Morning stiffness" is one of the hallmark symptoms of rheumatoid arthritis. While many people with other forms of arthritis have stiff joints in the morning, it takes people with rheumatoid arthritis more than an hour (sometimes several hours) before their joints feel loose.
  • Swelling - Fluid enters into the joint and it becomes puffy; this also contributes to stiffness.
  • Pain - Inflammation inside a joint makes it sensitive and tender. Prolonged inflammation causes damage that also contributes to pain.
  • Redness and warmth - The joints may be somewhat warmer and more pink or red than neighboring skin.
  • Shortness of breath or chest pain - Because rheumatoid arthritis can affect the blood vessels and muscle of the heart, people with the condition are at a greater risk for heart attacks and heart failure.
  • Numbness or tingling - Rheumatoid arthritis can cause connective tissues in the hand or foot to become inflamed and push up against a nerve causing numbness and tingling. As a result, you might experience weakness or clumsiness, or even nerve damage.
  • Inability to move or raise your hand or foot - Compared to numbness or tingling, suddenly not being able to raise or move a hand or foot is a much more severe complication to result from rheumatoid arthritis. It's also a much more rare occurrence, involving damage to nerves that are connected to muscles.
  • Spots on or around your fingertips - Little red or black spots on or around the fingernails may mean that the tissue in those areas has died from inflamed small blood vessels.
  • Red, inflamed eyes - Blood vessels in the eyes are another common target of rheumatoid arthritis, especially among people with more serious forms of the disease.
  • High fever and other signs of infection - Another problem from RA treatment is infection related to biologic drugs such as Cimzia, , Humira, and Simponi.

The involvement of pain mediators; prostaglandin and 5-hydroxytryptamine in analgesic activity of Ashwagandha was studied by pretreatment with paracetamol (100 mg/kg, ip) and cyproheptadine (10 mg/kg, ip). The analgesic activity of Ashwagandha was potentiated significantly by cyproheptadine, however, paracetamol failed to exhibit any significant change in its activity, suggesting the involvement of serotonin, but not prostaglandins in the analgesic activity of Ashwagandha.
